The bus was on route from the port city of Karachi to Sukkur when it hit the tanker near Nooriabad on super highway between Karachi and Hyderabad late Saturday.
The victims burned to death when both vehicles caught fire.
Several women and children were reportedly among the fatalities. Some passengers managed to escape the blaze, with severe burns.
Officials say all bodies have now been pulled out of the bus, but are difficult to identify.
The oil tanker was reportedly loaded with 40,000 liters of diesel fuel which caused the heavy fire.
